Histone Deacetylases (HDAC) & Histone Acetyltransferase (HAT)   

Eukaryotic DNA is highly organized and packaged into the nucleus. The organization and packaging are achieved through the addition of proteins, including core histones H2A, H2B, H3 and H4, which form a complex structure, the chromatin, together with DNA. The modification of core histones is of fundamental importance to conformational changes of the chromatin. The level of acetylation is related to transcription activity, and then the acetylation induces an open chromatin confirmation that allows the transcription machinery access to promoters. Histone deacetylase (HDAC) and histone acetyltransferase (HAT) are enzymes that influence transcription by selectively deacetylating or acetylating the ε-amino groups of lysine located near the amino termini of core histone proteins. Chromatin acetylation correlates with transcriptional activity (euchromatin), whereas deacetylation correlates with gene silencing. HDACs are also involved in the reversible acetylation of non-histone proteins. Altered HDAC and/or HAT activities are present in many types of cancers.

Mammalian HDACs have been classified into three classes. Class I (HDACs 1, 2, 3 & 8; each of which contains a deacetylase domain exhibiting from 45% to 93% identity in amino acid sequence) are homologs of yeast RPD3 and localize to the nucleus. Class II (HDACs 4, 5, 6, 7, 9 & 10) are homologs of yeast Hda1 and are found in both the nucleus and cytoplasm. The molecular weights of which are all about twofold larger than those of the class I members, and the deacetylase domains are present within the C-terminal regions, except that HDAC-6 contains two copies of the domain, one within each of the N-terminal and C-terminal regions. HDAC11 has properties of both class I and class II HDACs. Class III (Sirt1 - Sirt7) are homologs of yeast Sir2 and form a structurally distinct class of NAD-dependent enzymes found in both the nucleus and cytoplasm. 

Conserved from yeast to human, HDAC classes I and II are inhibited by trichostatin A (Prod. No. 380-068) and appear to use a divalent zinc-binding motif. The metal-coordinated active site activates an H2O molecule for direct targeting and hydrolysis of the acetyl group to form acetate. Acetylation of lysines in histones neutralizes the positive electric charge between the negatively charged DNA backbone and tips the balance towards relaxing the chromatin, while deacetylation would shift the balance back to condensing the chromatin and silencing gene expression. In a similar way PARP-1 adds to histones hundreds of negatively charged ADP-ribose units, which repel histones away from the negatively charged DNA backbone and thus induces chromatin relaxation to facilitate accession of DNA repair enzymes and gene expression. 

Unlike HDAC classes I and II, Sir2-like proteins catalyze a unique reaction, that like PARP-1, requires the coenzyme NAD+. In this reaction, nicotinamide is liberated from NAD+ and the acetyl group of the substrate is transferred to cleaved NAD+, generating the novel metabolite O-acetyl-ADP ribose. Due to the NAD+ dependency, Sir2 might be the link between metabolic activity and genome stability and, finally, aging.

Inhibitors of HDAC classes I and II emerge as potent anti-cancer agents. A proposed mechanism for the anti-tumor effects of HDAC inhibitors is that the accumulation of acetylated histones leads to activation (and repression) of the transcription of a selected number of genes whose expression causes inhibition of tumor cell growth and induction of apoptosis. 

Disruption of HDACs has been linked to a wide variety of human cancers. HDAC inhibitors have been shown to be potent inducers of growth arrest, differentiation, and/or apoptotic cell death. Some newly synthesized compounds are potentially effective agents for cancer therapy and, possibly, cancer chemoprevention.

The histone acetyltransferase assay kit is a fluorescent method to assay samples for histone acetyltransferase (HAT) activity or to screen HAT inhibitors. The HAT Assay Kit includes enough recombinant human p300 to screen 96 wells of inhibitor compounds, and anacardic acid is included for use as a control, as it is a potent HAT inhibitor. All assay buffers have been thoroughly tested to ensure that no components interfere with either activity or inhibition assays. Histone H3 and Histone H4 N-terminal peptides are included as a HAT substrates, so you can use the one best suited for your HAT enzyme.

HAT Activity Assay Kit

Histone acetyltransferases (HATs) have been implicated to play a crucial role in various cellular functions, such as gene transcription, differentiation, and proliferation. BioVisions HAT Activity Colorimetric Assay Kit offers a convenient, nonradioactive system for a rapid and sensitive detection of HAT activity in mammalian samples. The kit utilizes active recombinant HAT as a positive control and acetyl-CoA as a cofactor. Acetylation of peptide substrate by active HAT releases the free form of CoA which then serves as an essential coenzyme for producing NADH. NADH can easily be detected spectrophotometrically upon reacting with a soluble tetrazolium dye. The detection can be continuous and suitable for kinetic studies. The kit provides a simple, straightforward protocol for a complete assay.
