In mammals, germ-line encoded pattern recognition receptors (PRRs)
detect the presence of pathogens either directly through recognition of
pathogen-associated molecular patterns (PAMPs) or indirectly through
sensing host-derived danger signals (DAMPs). The innate immune system
comprises several classes of PRRs that allow the early detection of
pathogens at the site of infection.
The membrane-bound toll-like receptors (TLRs) detect microbes on the
cell surface and in endosomes. The RIG-1 (retinoic acidinducible gene
I)-like receptors (RLRs; RLHs) and the Nod-like receptors (NLRs;
nucleotide-binding domain leucine-rich repeat containing receptors)
detect microbial components in the cytosol.

The NAD+ biosynthesis pathway in mammals consists of two steps involving
the enzymes nicotinamide phosphoribosyltransferase (NAMPT [EC
2.4.2.12.]) and nicotinamide/nicotinic acid mononucleotide
adenylyltransferase (Nmnat). NAMPT converts nicotinamide into
nicotinamide mononucleotide (NMN) which then is converted to NAD+ by
Nmnat. The rate limiting enzyme in the biosynthetic pathway of NAD+ is
NAMPT. Originally, NAMPT was identified as pre-B cell colony enhancing
factor (PBEF) a presumptive cytokinelike protein involved in subclinical
inflammation. Later the protein was renamed visfatin, a visceral
fat-derived adipocytokine believed to mimic insulin action. However this
report was controversial resulting in the retraction of the paper.
Although NAMPT may not directly bind to the insulin receptor as
originally thought, NAMPT does regulate insulin secretion in β cells and
can function as a cell survival factor. |

In 1996 the Toll protein in Drosophila was discovered to be important
for defense against microbial infection. One year later a human homolog,
now known as Toll-like receptor 4 (TLR4), has been identified through
database searches. To date, 13 mouse and 10 human TLRs have been
identified. TLRs induce signaling pathways which result in a variety of
cellular responses that include the production of interferons (IFNs) and
various cytokines, such as tumor necrosis factor (TNF)-α, interleukin
(IL)-6 and IL-12. The cytoplasmic region of TLRs consists of a Toll/IL-1
receptor (TIR) domain, showing high similarity to that of the IL-1
receptor family. The extracellular region mainly consists of
leucine-rich repeats (LRRs) which are important for the recognition of
PAMPs. |
