The family of Toll-like receptors (TLRs) plays key roles in pathogen recognition and regulation of the innate immune response. Toll-like receptor 3 (TLR3) has been shown to specifically recognize double-stranded RNA, a molecular pattern associated with viral infection. TLR3 forms a complex with the adaptor molecules TRIF and TBK that results in the phosphorylation of IRF3. The activated IRF3 translocates to the nucleus and along with other transcription factors like NF-κB up-regulates the production of IFN-β. TLR3 is primarily expressed inside the cell associated with the endosomes. Thus, signalling by TLR3 requires intracellular delivery of dsRNA. Today the most widely used dsRNA is the synthetic polyinosinic:polycytidylic acid [poly(I:C)] due to its potent activity and increased stability.
