BBQ
Assays - Principle
The BBQ Assay is a particle based immunoassay for the quantitative detection of cytokines and other cell communication factors (like those involved in apoptosis). Antibody-coated particles in the close environment of cells bind to the antigen before any dilution into the supernatant occurs. The protein bound to the particles is now stained with a second, fluorescence-labelled antibody forming a sandwich layer and the particles are analysed by flow cytometry. The fluorescence shift of the particles is proportional to the amount of protein trapped by the particles. The ease of use of the magnetic particles enables the detection of protein in the presence of cells. |

Advantages:
No enzyme reaction involved: Intra-
and interassay variation is very small. Considerably less washing steps
are required than in ELISA. Wider detection range makes dilution
of the samples unnecessary.
Extremely flexible: Sample
volume, cell number and incubation time can be varied to achieve the
desired detection range.
Detects signals from various matrices: Cell
culture media, sera and plasma can be used.
Conservation of the signal: Since
the antibodies are covalently bound to the particles (not adsorbed as on
an ELISA plate), the incubation and storage time of the antigen
loaded particles is only limited by the stability of the antigen.
Quick and easy handling: Magnetic
particles make washing very easy, no relevant loss of particles occurs.
Multiparameter analysis possible: Up
to 3 analytes per sample can be measured in one run. General
